Therapeutic drug conjugated with nanoparticle for targeting delivery has rapidly gained interest in the area of nanomedicine. The nanocarriers, such as quantum dots (QD), are usually internalized into tumor cells through endocytic pathway. However, the study of nanoparticle endocytosis presents much challenge to conventional tools. Herein we use total internal reflection fluorescence flow cytometry (TIRF-FC) to investigate the process of QD uptake by cells at the single cell resolution. The kinetics of that process is also studied that the uptake speed through cell plasma membrane is a constant of 5.5 nm/min.
